PHAME NANO
Traditionelle Biopsie ist derzeit der Goldstandard in der Histopathologie. Optische Biopsie ermöglicht jedoch schnellere und genauere Diagnosen. PHORNANO ist ein österreichisches Startup, welches den Fokus auf Nanomaterialien und deren Anwendung in der klinischen Diagnose hat. Gemeinsam mit RECENDT, einem österreichischen Forschungsinstitut mit großer Erfahrung in biomedizinischer Bildgebung und mit CETENE, einem brasilianischen Forschungsinstitut welches sowohl große Erfahrung in der biomedizinischen Bildgebung sowie mit Nanomaterialien hat, zielt PHORNANO darauf ab, einen wichtigen Schritt auf den Gebieten Nanomaterialien und klinische Diagnostik zu machen.

Ziel des Projektes ist es, neue Nanomaterialien zu entwickeln, welche als multimodale „Markierungen“ für die Standard-Fluoreszenzmikroskopie, die STED-Mikroskopie, sowie für die photoakustische Mikroskopie geeignet sind. Die Ziele des Projekts sind insbesondere:
• Entwicklung von „Nano-Markierungen“, basierend auf metallischen Nanopartikeln und Halbleiter-Quantenpunkten.
• Funktionalisieren dieser Nanopartikel und Quantenpunkte um das Markierung von Zellen und Zellstrukturen zu ermöglichen.
• Biologische Bildgebung auf zellulärer und molekularer Ebene mittels Fluoreszenzmikroskopie und photoakustischer Mikroskopie.
• Charakterisierung der Nanopartikel und Quantenpunkte und Bewertung ihrer Anwendbarkeit für die photoakustische Mikroskopie, Fluoreszenzmikroskopie und STED-Mikroskopie.
• Ersetzen der konventionellen molekularen Fluorophore durch diesen neuartigen Nanomaterialien und Untersuchung von unerwünschten Effekten, welche in das biologische System eingebracht werden.

Dieses Pilotprojekt dient auch als Ausgangspunkt für die Entwicklung einer Bioimaging-Plattform auf Basis multimodaler Bildgebung unter Verwendung dieser neuartigen Nanopartikeln.

Abstract

PHAME NANO
Conventional biopsy is the gold standard in histopathology. Optical biopsy (based on bio-imaging) potentially leads to a much faster and more accurate diagnosis. PHORNANO is a startup company with a strong focus on nanomaterials and its applications in clinical diagnostics. Together with RECENDT, an Austrian research institute with profound knowledge in bio-imaging, and with CETENE, a Brazilian research institute with profound knowledge in bio-imaging as well as in nanomaterials, PHORNANO aims to make a significant step into the field of nanomaterials as well as clinical diagnostics.

The goal of the project is to develop new nanomaterials which are suited as multimodal labels for standard fluorescence microscopy, STED microscopy, and photoacoustic microscopy. Particular goals are to:
• Develop nano-labels, based on metallic nanoparticles and semiconductor quantum dots.
• Functionalize these nanoparticles and quantum dots to allow labelling of cells.
• Perform bio-imaging at cellular and molecular level by means of fluorescence microscopy and photoacoustic microscopy.
• Characterize the nanoparticles and quantum dots and assess their applicability to photoacoustic microscopy, fluorescence microscopy, and STED microscopy.
• Substitute conventional molecular fluorophores with novel nanomaterials and investigate undesired effects introduced to the biological system.

This pilot project shall serve as a starting point for the development of a bio-imaging platform based on multi-modal imaging using novel nanoparticles with an emphasis on clinical diagnostics.
