Lefamulin – Therapieoption für COVID-19 Patienten

Projektbeschreibung

Neben der potenten antibakteriellen Aktivität zeigen Antibiotika aus der Klasse der Pleuromutiline, und allen voran das in Wien erfundene und in Europa, Kanada und den USA bereits zugelassene Antibiotikum Lefamulin (Xenleta™), auch eine antivirale und entzündungshemmende Aktivität. Xenleta™ ist derzeit für die Behandlung von bakterieller Lungenentzündung als Tablette und als Infusion kommerziell erhältlich (https://www.xenleta.com/). Weitere antibakterielle Anwendungsbereiche sind in Entwicklung. Nabriva wird im Zuge des vom FFG geförderten Projekts „Lefamulin&COVID-19“ Lefamulin auf seine antivirale und entzündungshemmende Aktivität näher untersuchen. Des Weiteren werden verschiedenste Pleuromutilin-Verbindungen aus Nabrivas umfangreicher Substanzbibliothek evaluiert. Positive Studienergebnisse könnten neue Wege zur Entwicklung innovativer Therapien für Viruserkrankungen einschließlich Coronaviren, wie z.B. COVID-19, eröffnen. Vorrangiges Ziel ist es, ein Datenpaket zu erstellen, welches die baldige Anwendung von Lefamulin an mit SARS-CoV-2 infizierten Personen zur Verringerung der Infektionsdauer bzw. Hospitalisierungsrate ermöglicht. Ferner soll eine Pipeline von neuen innovativen antiviralen und/oder entzündungshemmenden Pleuromutilinen entwickelt werden.

Abstract

Antibiotics from the class of the pleuromutilins, and particularly lefamulin (Xenleta ™), have demonstrated initial antiviral and anti-inflammatory activity in addition to the known potent antibacterial activity. Xenleta ™ was invented in Vienna, is approved in Europe, Canada and the USA, and is commercially available as a tablet and infusion for the treatment of bacterial pneumonia (https://www.xenleta.com/). Further antibacterial indications for Xenleta™ are under development. As part of the FFG-funded project “Lefamulin & COVID-19” Nabriva will investigate lefamulin for its antiviral and anti-inflammatory activity in more detail. In addition, various pleuromutilin compounds of Nabriva's comprehensive and unique substance library will be evaluated. Positive study results could trigger the development of new treatment options for viral diseases including coronaviruses e.g. COVID-19. Primary goal is to compile robust data enabling the use of lefamulin for the treatment of patients infected with SARS-CoV-2 for the reduction of severity and duration of infection. Furthermore, new innovative antiviral and anti-inflammatory pleuromutilins will be developed to enter the pipeline.
